Provided is an IL-I7 binding antibody or functional fragment thereof comprising both heavy (VH) and light chain (VL) variable domains; wherein said IL-I 7 binding antibody or functional fragment thereof comprises at least one antigen binding site comprising: a) an immunoglobulin heavy chain variable domain (VH) which comprises in sequence hypervariable regions CDRI, CDR2 and CDR3, with specified sequences; and b) an immunoglobulin light chain variable domain (VL) which comprises in sequence hypervariable regions CDRI', CDR2' and CDR3', said CDRI ' having specified sequences capable of inhibiting the activity of 1 nM human IL-I7 at a concentration of less than 5 nM by 50%, said inhibitory activity is measured on IL-6 production induced by hu-IL-I7 in human dermal fibroblasts, and wherein CDR sequences are according to the Kabat definition. Also provided are variants of the antibodies and corresponding DNA constructs, expression vectors and processes of production as well as uses of the antibodies for the manufacture of medicaments.
