Transgenic ungulates having reduced prion protein activity and uses thereof

Provided is a viable bovine or a bovine cell comprising a non-naturally occurring mutation in one or both alleles of an endogenous prion nucleic acid, wherein said mutation comprises an insertion of a transcription termination sequence and/or STOP codon into a prion nucleic acid. Also provided are bovines comprising a prion gene mutation that reduces the expression of an endogenous antibody. Further provided are bovines and bovine cells which also comprise a positive selection marker.
