A mutant strain of Amycolaptosis orientalis is provided to improve preparation yield of vancomycin by culturing the mutant strain in a medium containing appropriately regulated nitrogen source and specific trace elements such as melanin biosynthesis inhibitor. A mutant strain of Amycolaptosis orientalis BNG 716(KCCM-10836P) produces vancomycin with higher purity and yield, and is isolated by treating a wild strain of Amycolaptosis orientalis ATCC-19795 with NTG(N-methyl-N'-nitro-N-nitrosoguanidine), selecting strains having enhanced self-tolerance against vancomycin, inducing mutation of selected strains with NTG, and selecting the strain showing maximized vancomycin producibility. A method for producing vancomycin hydrochloride comprises the steps of: (i) seed-culturing Amycolaptosis orientalis BNG 716(KCCM-10836P); (ii) culturing the seed-cultured mutant strain in a culture medium containing 12-18% of dextrin, 2.2-3.8% of bean flour, 1.9-2.9% of potato protein, 0.10-0.14% of sodium chloride, a trace amount of minerals and vitamin, and a regulated concentration of melanin biosynthesis inhibitor; (iii) filtering the cultured medium solution with a microfilter to remove the mutant strain and obtain crude vancomycin; and (iv) subjecting the crude vancomycin to cation exchange resin column, anion exchange resin column, hydrophobic adsorption resin column, activated charcoal and activated alumina adsorption column, and crystallizing the purified vancomycin with hydrochloric acid.
