<P>PROBLEM TO BE SOLVED: To provide a double-stranded RNA having high nuclease resistance and exhibiting an excellent RNA interference effect. <P>SOLUTION: This double-stranded RNA having a sense chain containing a base sequence complementary to a target sequence in a target gene and an antisense chain containing a base sequence complementary to the sense chain, and restricting the expression of the target gene is made so as to be equipped with the following (i) to (iv) constitutions as one unit. (i) A dangling end is made as being equipped at only one of the 3'terminal of the sense chain or the 3'terminal of the antisense chain. (ii) At the side without having the dangling end, the sense chain and antisense chain are completely complementary paired to form a smooth terminal. (iii) At the sense chain or antisense chain, a nucleotide at first to 6th from the smooth terminal end side may be constituted with a deoxyribonucleotide. (iv) In the case that the dangling end is formed at the 3'terminal of the sense chain, the antisense chain is constituted with 23 bases, and in the case that the dangling end is formed at the 3'terminal of the antisense chain, the antisense chain is constituted by 23 to 26 bases. <P>COPYRIGHT: (C)2008,JPO&INPIT |
