A method for preparing a tire tread rubber composition is provided to improve mechanical properties remarkably and to enhance frictional force and low heat build-up. A method for preparing a tire tread rubber composition comprises the steps of mixing 20-80 parts by weight of a layered silicate/rubber nanocomposite, 20-80 parts by weight of a rubber material and a reinforcing agent; and melting, mixing and curing the mixture by using an internal mixer under the condition of an initial temperature of 60-120 deg.C, a shear velocity of 40-200 /sec and a shear stress of 0.75x10^9 to 2.5x10^9 kJ/kg for 100-1,000 sec.
