METHOD FOR SCREENING PHARMACEUTIC AGENTS AGAINST METABOLIC DISORDER AND SKIN DISORDER BY USING PEROXISOME PROLIFERATOR ACTIVATED RECEPTOR (PPAR delta;/beta;) AND PPAR CO-ACTIVATORS

A method for screening pharmaceutical agents for treating metabolic disorder and skin disorder by using a peroxisome proliferator activated receptor(PPAR delta/beta) and PPAR co-activators is provided to screen a large quantity of candidate compounds simultaneously, and reduce the screening costs by detecting binding between PPAR delta/beta to PPAR co-activators. A method for screening pharmaceutical agents for treating metabolic disorder and skin disorder by using the peroxisome proliferator activated receptor (PPAR delta/beta) and PPAR co-activators comprises the steps of: (1) coating the PPAR co-activators on a plate; (2) adding a candidate compound together with PPAR delta/beta protein into the plate; (3) binding an antibody against PPAR delta/beta; (4) reacting a secondary antibody labeled with an enzyme capable of binding to the antibody against PPAR delta/beta; and (5) detecting the labeled secondary antibody, wherein the PPAR co-activator is selected from steroid receptor co-activator SRC-1, transcription mediator TIF-2 or p300 protein and the metabolic disorder is obesity, diabetes, hypertension, cardiovascular disease or hyperlipidemia.
