Immunological determination of biomolecules (I) comprises performing immunological reaction of (I) in a liquid system. Immunological determination of biomolecules (I) by performing immunological reaction of (I) in a liquid system comprises: (A) labeling (I), before reaction, with an imidazole derivative (II) or an enzyme (III); (B) adding a trigger substance (IV) to the liquid system so as to cleave protecting groups and releasing (by enzyme-catalyzed or direct hydrolysis) a colorless tetraamino-biimidazole (V) which is immediately converted, by oxygen in the air, to a colored tetraazafulvene (VI) and one equivalent of hydrogen peroxide; (C) quantification of hydrogen peroxide by a high-sensitivity chemiluminescent method; and (D) quantitative determination of (I) from the result.
