An attenuated Pasteurellaceae bacteria comprising a mutation in a gene having a nucleotide sequence set forth in SEQ ID NO:76 is described. The mutation results in decreased yiaO biological activity of a gene product encoded by the mutated gene, wherein the decreased yiaO biological activity attenuates the bacteria. Also described are vaccine compositions comprising the bacteria, isolated polynucleotides and polypeptides, vectors, non-human host cells, antibodies and methods for producing a gram-negative mutant. A method of identifying an anti-bacterial agent comprises the steps of assaying potential agents for the ability to interfere with expression or activity of a yiaO gene product, represented by the amino acid sequence set forth in SEQ ID NO: 77, and identifying an agent that interferes with yiaO gene expression or biological activity of the gene product.
