A primer amplifying the base sequence in a target region selected from among a part of the D2 domain in an rRNA gene of a yeast belonging to the genus Saccharomyces and its complementary strand, characterized by containing: (a) as a first segment, a base sequence annealing to the rRNA gene of the yeast belonging to the genus Saccharomyces and acting as a primer; and (b) as a second segment, a base sequence being complementary with the 3'-side base sequence of the first segment and located in the 5'-side of the first segment.
