PROBLEM TO BE SOLVED: To provide a new method for testing antibacterial activity, etc., by using an OS-1 gene, etc.
SOLUTION: A transformed cell is given by introducing a gene of an osmotic pressure-sensitive histidine kinase without a cell membrane penetrating region into a cell from which at least one or more hybrid sensor kinases are deleted in such a form that the gene functions. The method for testing the antibacterial activity of a substance has a first process for culturing the transformed cell in the presence of a substance to be tested, a second process for measuring an amount of an intracellular signal transmitted from the osmotic pressure-sensitive histidine kinase without the cell membrane penetrating region which is expressed in the transformed cell cultured in the first process, or measuring an index value having correlation with the amount of the intracellular signal, and a third process for evaluating the antibacterial activity of the substance to be tested based on a difference of the amount of the intracellular signal measured in the second process from a control, or a difference of the index value having the correlation measured in the second process from the control.
COPYRIGHT: (C)2005,JPO&NCIPI |
