PURPOSE: Provided is a Bacillus sp. strain and protease extracted therefrom which is very stable to negative ionic surface active agent, oxidant and organic solvent, and effectively used for various industrial purposes, such as detergents for clothes or contact lenses, processing materials for foods, feedstuffs, fabrics and natural skins, and disposals of organic waste. CONSTITUTION: A strain is Bacillus sp. 104(KCTC 10166BP). A method of extracting protease from the strain comprises the steps of: preparing a coenzyme solution by culturing Bacillus sp. strain, centrifuging it and recovering the upper-leveled culture solution; absorbing the coenzyme solution, and eluting active fractions of the coenzyme solution with a sodium phosphate buffer solution(pH7.5) containing 30% of acetone; adding the eluate to the chromatography column, and eluting active fractions thereof with a 20mM tris buffer solution(pH8.2); adding the eluate to negative ion exchange chromatography and positive ion exchange chromatography, cleaning it with a buffer solution, and eluting active fraction thereof with a 40mM sodium phosphate(pH6.0); and isolating protease with a molecular weight of 28kDa therefrom.
