PURPOSE: An enzymatic preparation method of optically active 2-hydroxy-4-phenylbutyronitrile and esters thereof in non-solvent phase is provided, thereby easily and environment-friendly preparing the compound, and reducing the preparation costs by repeatedly reusing an acyl donor and catalyst. CONSTITUTION: The enzymatic preparation method of optically active 2-hydroxy-4-phenylbutyronitrile and esters thereof in non-solvent phase comprises the steps of: dissolving racemic 2-hydroxy-4-phenylbutyronitrile in acyl donor in non-solvent phase; and esterification of the racemic 2-hydroxy-4-phenylbutyronitrile dissolved solution, wherein the acyl donor is vinylester compound, organic acid anhydride, or acetateisoprophenyl.
