Genetically modified penicillin amidase and methods of producing it

Penicillinamidase (I) comprising at least one peptide unit (PU) to which (part of) a linker peptide (LP) is bonded so that its activity is increased, relative to an enzyme containing the same PU but not linked to LP, is new. Independent claims are also included for the following: (1) pro-penicillinamidase (II) having an LP that links first and second PU, where LP has a sequence that delays or suppresses cleavage of LP from the first PU; (2) nucleic acid (III) that encodes (I) or (II); (3) altering the activity of an enzyme (E) by modifying a precursor that includes an LP linking two PU such that complete cleavage of LP from the first PU is delayed or inhibited; (4) preparing 6-aminopenicillanic acid (6APA) and/or 7-aminodeacetoxycephalosporanic acid (7ADCA) using (I) or (II); (5) composition that contains at least one of 21 specified oligopeptides (III); and (6) vector or host cell containing at least one (III).
