PURPOSE: A human histone protein-producing plant cell and a preparation method thereof are provided. The provided human histone protein is fully modified and has activity, so that it can be useful for treatment of rheumatoid arthritis. CONSTITUTION: A method for preparing a human histone protein-producing plant cell comprises the steps of: amplifying human histone H1.5 gene from cDNA library of human placenta by PCR; inserting the human histone H1.5 gene into a vector pCAM; transforming Agrobacterium EHA105 with the recombinant expression vector; and co-culturing the transformed Agrobacterium EHA105 together with Nicotiana tabacum L. cv. Bright yellow 2(BY02).
