Protein mixtures are analysed by (a) enzyme digestion of the protein mixture to form digestion peptides then <SL> <LI>b) separating the mixture of digestion peptides into chromatographic fractions; <LI>c) collecting the individual chromatographic fractions; <LI>d) preparing samples on a sample support, where the samples contain the digestion peptides from the respective chromatographic fractions and a matrix substance; <LI>e) ionizing the digestion peptides from the samples by matrix assisted laser desorption in a time-of-flight mass spectrometer to form digestion peptide ions; <LI>f) determining the masses of the digestion peptide ions from the times of flight of the said ions; <LI>g) measuring daughter ion spectra for all or for selected digestion peptide ions from all or from selected samples by MALDI tandem time-of-flight mass spectrometry; and <LI>h) identifying associated proteins by searching in protein sequence, cDNA, DNA or EST databases. </SL> Protein mixture is e.g. a proteome. |
