<p>The invention relates to a single-cell regeneration process for plants of the Agave tequilana Weber var. azul species using in vitro indirect somatic embryogenesis. The invention relates to plants generated from a single cell originating from dedifferentiated tissue (callus). The inventive process comprises the following steps: introducing somatic cells into a culture medium which is prepared using the basic salts of the MS (Murashige and Skoog, 1962) medium and L2 vitamins (Phillips and Collins, 1979); the medium is gelled with agar, using sucrose as a carbon source, and supplemented with 2,4-dichlorophenoxyacetic acid auxin and cytokinin benzyladenine. The exposure time to the induction medium is between 15 and 80 days. Once the induction period is finished, the embryogenic callus are transferred to a solid culture medium for expression and germination. The plant material is incubated under the following conditions: temperature of 27+/-°C; light intensity of 1,500 lux; and a photoperiod of 16 hours. The inventive method can be used for in vitro genetic improvement and micropropagation of the species.</p> |
