PROBLEM TO BE SOLVED: To provide a new method for linking a single-stranded RNA and a single-stranded DNA with each other, for example, to provide a new method for constructing in vitro virus genome. SOLUTION: This method for producing an RNA-DNA conjugate comprises the steps of (1) annealing a single-stranded RNA and a single-stranded DNA or a derivative thereof having sequences complementary to each other and (2) treating the annealed product with an RNA ligase to link the 3'-terminal of the single-stranded RNA and the 5'-terminal of the single-stranded DNA or the derivative thereof with each other. |
