A method of constructing a DNA mass into which deletion mutation has been transferred stepwise from the 5'- or 3'-terminal side by using homologous recombination; the DNA mass thus constructed; a yeast to be used in this method; and a method of identifying a protein-binding region by using the above method and the two-hybrid method.
