Polynucleotide sequence capable of directing translation initiation in a plant expression system in a cap-independent manner, wherein the polynucleotide sequence directs translation initiation from an AUG codon. In particular, the invention relates to a polynucleotide sequence which comprises the 5' UTR of Rhopalosiphum padi virus (RhPV) which comprises an internal ribosome enty site (IRES).
