<p>A process for producing isoelectrically homogenized Fab' antibody characterized by involving: the first step of providing an Fd chain gene encoding the VH region and the CH1 region of Fab' antibody; the second step of site-specifically mutating at least one codon encoding an amido-containing amino acid residue in the CH1 region of the above Fd chain gene into another codon encoding an amido-free amino acid residue other than cysteine to thereby give a modified Fd chain gene; the third step of ligating the modified Fd chain gene with the L-chain gene encoding the L-chain of the above-described Fab' antibody in such a manner as allowing the expression to thereby give a modified Fab' antibody expression gene; and the fourth step of transforming a host cell with the above-described modified Fab' antibody expression gene and culturing the thus obtained transformant to thereby give the isoelectrically homogenized Fab' antibody.</p> |
