PURPOSE: A method for detecting the sequence variation of nucleic acids by shifted termination analysis(STA) is provided, which is more cost- and time- effective but less labor intensive than a conventional method. CONSTITUTION: The method for detecting the sequence variation of nucleic acids by shifted termination analysis(STA) comprises the steps of: a) preparing a primer complementary to the upper stream of a target nucleotide sequence at a predetermined site on the template of an interested nucleic acid; b) preparing an unpaired nucleotide by treating a sample containing the interested nucleic acid provided that the nucleic acid is double-stranded, or using the interested single-stranded nucleic acid itself, for the step (c); c) annealing the primer to the target nucleic acid to produce a double-stranded primer-nucleic acid; d) mixing the double-stranded primer-nucleic acid with primer extension reagents; e) carrying out enzymatic or chemical primer extension reaction; and f) detecting labeled non-terminators.
