PROBLEM TO BE SOLVED: To reduce the amount of mobile phase consumption, reduce a load pressure, and achieve high-speed separation by forming a silica gel in double- pore structure where a silica skeleton with a pore with each specific size in a capillary whose inner diameter is equal to or less than a specific value is interwined. SOLUTION: A compound that is thermally decomposed is dissolved in a reaction solution in advance, a gel consisting of a solvent-rich phase and a skeleton phase that is rich in silica and has a pore on the surface is adjusted in a capillary, and the gel in wet state is heated, thus thermally decomposing the compound and drying the gel. As a result, in a column, a silica gel in double pore structure where a silica skeleton with a pore (through pore) with 0.5-5 μm size and a pore (meso pore) with 2-50 nm size in a capillary whose inner diameter is 1,000 μm or less is interwined is formed. In this case, when the diameter of a through pore is smaller than 0.5 μm, a load pressure becomes larger. On the other hand, when the diameter is larger than 5 μm, a separation performance becomes inferior. Also, when the diameter of the meso pore is smaller than 2 nm, solvent dispersion becomes difficult. On the other hand, when it is larger than 50 nm, separation becomes insufficient. |
