PURPOSE: To effectively modify a keratin protein fiber product and produce a large amount of a protease specific to the keratin protein at a low cost. CONSTITUTION: This protease NH has the following enzymological properties. (1) The enzyme hydrolyzes various proteins and produces oligopeptides or amino acids. (2) It has high activity against soluble proteins such as casein and insoluble proteins such as keratin. (3) The optimum pH is 7.0-9.0 for the reaction at 37 deg.C using casein as a substrate. (4) It is extremely stable within the pH range of 4.5-9.5 by the treatment at 55 deg.C for 20min at various pH values using casein as a substrate. (5) The optimum temperature is 50-60 deg.C for the reaction at pH 7.5 using casein as a substrate. (6) The residual activity is >=90% in the case of treating at pH 7.5 and 40 deg.C for 10min using casein as a substrate or treating at 50 deg.C for 10min after addition of 5mM of Ca<2+> . |
