发明名称 GENE POSITIVELY CONTROLLING TRANSFER OF YEAST MANNOSE-1-PHOSPHATE AND PRODUCTION OF HIGH MANNOSE TYPE NEUTRAL SUGAR CHAIN USING DEFECTIVE VARIANT OF THE SAME
摘要 PROBLEM TO BE SOLVED: To obtain a gene capable of positively controlling the mannose-1- phosphate transfer of yeast coding specific amino acid sequence and giving a high mannose type neutral oligosaccharide or a glycoprotein in plenty and high purity by a genetically-engineered process of the yeast. SOLUTION: This new gene is capable of positively controlling the mannose-1- phosphate transfer of the yeast coding an amino acid sequence expressed by the formula, producing a high mannose type neutral oligosaccharide having a sugar chain structure same as that of human and a glycoprotein by adding the high mannose type neutral oligosaccharide chain to an aspartic acid residue in the protein, starting from the yeast by using the recombinant DNA technology. This new gene is obtained by extracting genomic DNA from Saccharomyces cerevisae, preparing the gene library by conventional method, cloning the gene by amplification using the primer consisting of a part of the gene coding the protein positively controlling the transfer of mannose-1- phosphate according to PCR method.
