PURPOSE:To obtain a creatine amidinohydrolase stable in high pH region and having small Km value. CONSTITUTION:This new creatine amidinohydrolase has the following physical and chemical properties. (a) Action, hydrolyzing 1mol of creatine to form 1mol of sarcosine and 1mol of urea; (b) substrate specificity, having substrate specificity to creatine; (c) optimum pH, 7-9; (d) optimum temperature, about 35-45 deg.C; (e) pH stability, stable at pH 5.0-10.5 by the treatment at 25 deg.C for 17hr; (f) thermal stability, stable up to about 45 deg.C by the treatment at pH 7.5 for 30min; (g) inhibitors, AgNO3, HgCl2, CuSO4, etc.; (h) molecular weight, about 80,000+ or -5,000 (gel-filtration). This invention also relates to a process for the production of the creatine amidinohydrolase. |
