PURPOSE:To obtain a new lambda phage DNA, useful for preparing a transgenic mouse and utilizable for a test on investigation of mutagenicity of a chemical substance for a gene by administering the test chemical substance to the mouse, preparing a cyclic plasmid from a DNA extracted from viscera thereof and using the resultant cyclic plasmid. CONSTITUTION:This lambda phage DNA has a base sequence integrated in the following order from the left arm to the right arm in a base sequence thereof: (1) an Hind III-recognition sequence; (2) a loxP sequence; (3) a sequence prepared by locating three sequences of an Escherichia coli gpt gene sequence, a base sequence having a plasmid replication origin and a chemical-resistant gene sequence according to a combination in an optional order; (4) the loxP sequence in the same direction as that of the loxP sequence and (5) the Hind III- recognition sequence. |
