The invention relates to a process for the detection and/or quantification of a nucleic acid sequence in cells or tissues by in situ PCR, characterised in that: - the membranes are permeabilised to the PCR reagents by treating the cells with a product for permeabilising and fixing the membrane under conditions of concentration and duration ensuring maintenance of the membrane structure, - the labelling of the amplification products of the PCR reaction is performed by incorporation of labelled triphosphate nucleotides, - where appropriate, a ligand specific for a membrane and/or cell constituent is added, the said ligand being labelled or capable of being labelled directly or via a chemical intermediate with a labelled conjugate.
