A method for determining the relative quantities of all cholesterol-containing lipoproteins in body fluids by fractionating the lipoproteins in an aliquot of the body fluid by electrophoresis on a support matrix and subsequently detecting them by an enzymatic treatment which comprises an incubation of the support material with cholesterol esterase and cholesterol dehydrogenase and leads to the formation of a detectable complex, and determining the relative quantities of the various lipoprotein classes, where the electrophoresis is carried out on a thin-layer matrix. The method makes it possible to determine HDL-, LDL-, VLDL- and LP (X)-cholesterol simultaneously in body fluids with high precision and at low concentrations. The electrophoresed thin-layer matrices obtained by the method according to the invention are, furthermore, easy to handle and store.
