<p>PURPOSE:To provide an expression plasmid for seed, containing a soybean seed storage protein glycinin gene promoter and a soybean seed storage protein glycinin gene terminator and useful for the investigation of the expression of the plant gene of seed. CONSTITUTION:Promoter region and terminator region separated from soybean seed storage protein glycinin gene GY1 are connected to both sides of a cloning site of plasmid pUC19 to construct expression plasmid pSUM-GY1. In the above construction process, the gene fragment of a promoter region of about 700 bp is amplified and separated by PCR method using the GY1 base sequence, a soybean genomic library is screened by using the gene fragment as a probe to obtain a gene fragment of about 4.4 kb containing complete GY1, subcloning the fragment, separating the promoter region and terminator region from the plasmid and connecting the regions to both sides of the cloning site of the plasmid pUC19 to obtain the objective expression plasmid pSUM-GY1.</p> |
