PRODUCTION OF COLORING MATTER OF MADDER USING HAIR ROOT

PURPOSE:To continuously and efficiently extract a coloring matter of madder from hair roots of madder. CONSTITUTION:A plant of madder is transformed with Ri plasmid possessed by Agrobacterium rhizogenes and hair roots produced by the transformation are cultured in a liquid medium. At this stage, the liquid medium is rotated and shaken to increase the concentration of dissolved oxygen. Then, the rotation and shaking are stopped to reduce the pressure of dissolved oxygen in the liquid medium. The liquid medium is adjusted to pH 7.0-9.0. The multiplication of the hair roots is stopped with rise in the concentration of dissolved oxygen and pH, and a coloring matter of madder is secreted in the liquid medium from the interior of the cells of the hair roots (extraction operation). The culture operation and extraction operation are alternately repeated to produce continuously and efficiently the coloring matter of madder by using the same selected clone.
