METHOD FOR OBTAINING RECOMBINANT SURFACE ANTIGEN OF HEPATITIS B, ANTIGEN AND VACCINE BASED ON IT

A recombinant surface antigen of hepatitis B is obtained by culture of an antigen producing strain such as Saccharomyces cerevisiae DAN-041/pBMK-1, deposited in collection VNIIG under No. Y-1857, and by biosynthesis of the antigen on a nutrient medium containing peptone and the yeast extract, while maintaining the non-organic phosphorous content in the nutrient medium in the course of the culture within the range of 40-200 mg/l and during biosynthesis, within the range of 5-50mg/l, with subsequent disintegration of the yeast cells and extraction of the antigen. In the course of the biosynthesis is provided an additional feeding of a fresh nutritient medium containing peptone, the yeast extract and glucose at the weight ratio of 0.05-0.15 : 0.15-0.15 : 1.0, respectively. A hepatitis B vaccine based on the recombinant surface antigen includes also an adjuvant, a preservative and a physiologically acceptable diluent.
