PURPOSE:To obtain a new polypeptide useful for activation of vascular endothelial cell, study of development mechanism of inflammation, serum diagnosis of thrombotic inflammation, clinical application, capable of specifically recognizing human GMP-140 having a specific amino acid sequence. CONSTITUTION:mRNA is isolated from PL7-6 hybridoma cell (FERM P-11,073) by a conventional procedure, cDNA is synthesized by the mRNA and amplified to select a clone containing a DNA encoding a variable range of heavy chain and light chain. The DNA is recovered, treated with a restriction enzyme, linked to a vector and transduced to a host to polypeptides of formula I (FR1 is polypeptide residue containing 23-28 amino acids; FR2 is polypeptide residue containing 14-16 amino acids; FR3 is polypeptide residue containing 30-34 amino acids; FR4 is polypeptide residue containing 9-11 amino acids; CDR1L is group of formula II; CDR2L is group of formula III; CDR3L is group of formula IV) and formula V (FR5 is polypeptide residue containing 29-36 amino acids; FR6 is polypeptide residue containing 10-16 amino acids; FR7 is polypeptide residue containing 32-25 amino acids; FR8 is polypeptide residue containing 12-14 amino acids; CDR1H is group of formula VI; CDR2H is group of formula VII; CDR3H is group of formula VIII). |
