PURPOSE:To provide a new protein originated from tissue or cell, effective for promoting the production of anti-DNA antibody, active in binding with DNA and useful as an antigen for the production of antibody for the diagnosis and treatment of autoimmune diseases, cancer, etc. CONSTITUTION:The objective protein capable of promoting the production of anti-DNA antibody and binding with DNA is produced by collecting KML1-7 cell line originated from an established lymphatic cell KML1 capable of producing and releasing a factor which promotes the production of anti-DNA antibody and is obtained from the total cell cluster of swollen lymph gland of MRL/1 mouse, culturing the cell line in a medium in the presence of serum until the system becomes confluent, collecting the proliferated cells by centrifugal separation, suspending the cells in a serum-free medium, mass-culturing in a CO2 incubator for 3 days, centrifuging the culture liquid to collect the supernatant, concentrating the supernatant by an ultrafiltration membrane, subjecting to gel filtration to collect active fraction, subjecting the fraction to anion exchange column chromatography, collecting the fraction eluted by >=0.2M of saline water and purifying the fraction by a heparin column and a reversed phase high- performance liquid chromatography. |
