PURIFICATION OF GRANULOCYTE COLONY STIMULATING FACTOR

A human granulocyte macrophage colony stimulating factor (GM- CSF) is purified from the culture medium of a recombinant DNA yeast by (a) concentrating the yeast culture medium with siprial ultrafiltration catridge, (b) precipitating the protein of the soln. at 4.7-5.0 by adding alcohol, and dissolving in the buffer soln. (pH 7.5), and (c) purifying the human GM-CSF contg. protein soln. by anion exchange chromatography using silica PAE 300 resin and hydrophobic chromatography using phenyl cephalose resin. The method is useful for the prodn. human GM-CSF in good yield and high purity.
