摘要 The process for the manufacture of very active and highly stabilised rigid G acylase- (aldehyde) support derivatives by multipoint covalent bonding between the amine groups of the enzyme and activated rigid supports containing a monolayer of identical aldehyde groups moderately separated from the surface of the said supports. Two main types of supports are used: i: inorganic solids (e.g. porous glass) and ii: synthetic polymers (e.g. epoxyacrylic resins). The process for the manufacture of penicillin G-acylase derivatives comprises three main stages: a: activation of the supports, b: immobilisation of the enzyme on the activated supports by strict control of all the variables which govern the intensity of the multiple enzyme-support interaction (concentration of aldehyde groups, pH, temperature, reaction time, etc.) and c: final reduction of the derivatives with sodium borohydride. In this way derivatives are prepared which have excellent mechanical properties and excellent catalytic activity and stability properties. The mechanical rigidity of these supports makes it possible to use these biocatalysts in fixed bed reactors with high flow velocities.
