NEW PLASMID AND PRODUCTION OF NADH OXIDASE USING THE SAME

PURPOSE:To obtain an NADH oxidase by synthesizing a plasmid holding a DNA region containing an NADH oxidase structure gene, a regulator gone region containing a promoter, etc., capable of controlling the expression of structure gene, etc. CONSTITUTION:A microorganism belonging to Escherichia coli K.12 is transformed with a plasmid containing (a) a DNA region containing an NADH oxidase structure gene, (b) a regulator gene DNA region containing a promoter and operator capable of controlling the expression of the above structure gene and (c) a DNA region containing a gene managing the autonomous proliferation of ColEI-type plasmid. The transformed microorganism is cultured in a culture medium and the objective NADH oxidase is separated from the cultured product. Plasmid pNOX-1 is a preferable example of the above plasmid.
