PURPOSE: To determine the concentration of a specimen in a sample by producing an enzyme by a quantity correlated with the concentration of the specimen, producing a product of a substrate acting on the enzyme and bringing the product into contact with a specific binder to be bound therewith. CONSTITUTION: At first, a solid phase,capturing system is prepared and a double conjugant of cumarin substrate-estradiol is produced on dextran-6-aminohexane. Subsequently, a solution of a dextran substrate is prepared followed by preparation of a conjugant of solid phase ouabain-antidigoxin-β-galactosides. A standard assay and an amplified assay are then performed using galactosides activity generated by pushing out a solutionlike digoxin.
