PURPOSE:To purify lymphotoxin (LT) useful as a carcinostatic agent in high yield, by treating a culture solution containing human LT by metallic chelate affinity chromatography. CONSTITUTION:Cell CHO 2-3E-4B strain producing LT is cultivated in a medium containing 2% bovine fetal serum to give a culture solution containing LT activity. Then the culture solution is charged to a column of chelating Sepharose 6B of affinity carrier chelated with Zn ion to adsorb LT on the carrier. Then the column is cleaned with a buffer solution containing NaCl, etc., a buffer solution having a linear concentration gradient such as imidazole, etc., is charged to the column, LT is eluted and purified. |
