PURPOSE:To decide fractions of substitution bodies handily, by measuring the amounts of 1, 2 and 3 carboxy methyl substituted glucoses formed by hydrolysis of carboxy methyl cellulose by uniform velocity electrophoresis. CONSTITUTION:A carboxy methyl cellulose sample is use as intact or regenerated by freezing dry, a mineral acid (e.g.: sulfuric acid) is used to hydrolyze (primary and secondary), neutralized and filtered to obtain a hydrolytic raw liquid. Then, the amounts of 1, 2 and 3 carboxy methyl substituted glucose and glucose (non-substituent) formed by hydrolysis in the raw liquid are measured by a uniform velocity electrophoresis and the mol ratio of each substituent is calculated. By this method, the fractions of the substitution bodies can be determined with a high reproducibility with a simple equipment in a short time. |
