Allergy-suppressing factor (ASF) is obtd. by (a) preparing hybridomas of human T cells able to express ASF, by the following operations:isolating human peripheral blood lymphocytes and cultivating them for 7 days with pokeweed mitogen, fusing with a clone of cellular Jurkat JKOT line resistant to thioguanine and ouabaine, and susceptible to hypoxanthine, aminopterine and timidine (HAT) by means of polyethylene glycol, cultivating the fused cells with rat timocytes as feed cells; (b) isolating the hybridomas by cultivation of individual clones in presence of penicillin and streptomycin, inserting the cultures in a human atmos. with 5% CO2 for 7 weeks in presence or absence of HAT; and (c) collecting the FSA produced from the supernatant liq. of the culture.
