<p>Cyclisation of the synthetic hetero-peptide phenoxy-acetyl L-cysteinyl D-valine is carried out using cell-free extracts of acremonium chrysogenum AS-C-80 with Isopenicillin N-synthesase activity. Incubation is in a medium contg. a mixt. of Tris-HCl 30mM, ditrotreitol 6mM, ferrous sulphate 0.1mM and sodium ascorbate 1mM, to regulate the pH at 7.8, at 20-37 deg.C for up to 120 mins.</p> |
