The process comprises (a) incubating free extracts of cells of Penicillium chysogenum AS-P-78 with Isopenicillin N-synthesase activity, in presence of phenylacetyl L-cysteinyl D-valine in a reaction medium of Tris-HCl buffer 30 mM, ditrotreitol (DTT) 6 mm, FeSO4 0.1 mM and sodium ascorbate 1 mm at pH 8-10 and 20-37 deg.C for 2 hrs. to cyclise the synthetic hetero-peptide and obtain Penicillin G.
