DNA AND ITS PREPARATION

PURPOSE:The m-RNA fraction is separated from macrophage which is obtained from a mammarian to which macrophage activator has been given and the fraction is used to form a DNA coding polypeptide with tumor necrosis factor (TNF) activity. CONSTITUTION:A macrophage activator such as lipopolysaccharide or BCG is given to the blood vessel (usually intravenously) of a mammarian, then macrophage is separated from liver, blood cells and lungs of the sensitized mammarian. Then m-RNA fraction containing m-RNA corresponding to polypeptide with TNF activity is separated and the DNA complementary for the fraction is prepared. The resultant C-DNA is inserted into cells of a microorganism by connecting it with a vector such as pBR322 and the microorganism is cultured to produce the polypeptide with TNF activity.
