PURPOSE:To recover and to purify desired protein efficiently, by cultivating a transformed bacterium in a medium containing a specific amount of an inorganic acid, and treating the formed bacterium by osmotic shock method. CONSTITUTION:A gene containing a structure gene of desired external protein is integrated into a vector which is a gene to code signal peptide and capable of linking the structure gene of desired external protein to just behind the downstream end of the gene to code the signal peptide and capable of multiplying in an expected host cell, to give a recombinant DNA capable of multiplying in the expected host cell. A Gram-negative bacterium is transformed with the recombinant DNA. The transformed bacterium is cultivated in a medium containing inorganic phosphorus in an amount to necessary to induce protein synthesis ability from the latter period of logarithmic growth phase to the former period of stopping phase. Then, the bacterium is treated by osmotic shock method, to collect a fraction containing desired external protein. |
