PURPOSE:To enable measurement of several kinds of enzyme activity with one measuring system and to simplify a titled device by measuring the decreased amt. of oxygen, etc. with one oxygen or hydrogen peroxide electrode and measuring the enzyme activity. CONSTITUTION:Metering pumps 8'', 8''' are operated to feed a sample soln. contg. enzyme from a sample liquid feed part 2 and a substrate-contg. liquid adjusted to obtain the conditions optimum for enzyme reaction from a substrate- contg. liquid feed part 3 respectively to a reaction part 1 for a prescribed amt. The reactive liquid obtd. by catalytic reaction is fed together with a pH adjusted buffer fed from a soln. feed part 7 by a pump 8' to the inside of an immobilized enzyme column 4. An electric signal is obtd. by the oxygen or hydrogen peroxide electrode incorporated into a flow cell 5. The immobilized enzyme column is of a cartridge type and if the immobilized enzyme column is exchanged, various enzyme activity can be measured and the need for laborious adjustment of the electrode is eliminated. Since the changing speed of the detecting material is measured, the influence of the electrode sensitive material incorporated preliminarily in the sample liquid is eliminated and the enzyme activity is measured with good accuracy. |
