摘要 PURPOSE:To obtain a carrier for an affinity chromatography which has a stable bond between a ligand and a base and uses, as the ligand, the mononucleotide having a high adsorptive allowance for a biopolymer, more particularly nuclease. CONSTITUTION:The 5'-deoxy-5'-aminonucleaside-2'(3')-phosphate expressed by the formula is brought into a coupling reaction with a base having a hydroxyl group subjected to an activation treatment with a cyan halide, thereby obtaining a carrier for an affinity chromatography. Such 5'-deoxy-5'-aminoliponucleoside- 2'(3')-phosphate is obtd. by bringing, for example, 5'-deoxy-5'-azide material into a reaction with a phosphorizing agent such as polyphosphoric acid, pyrophosphoric acid or the like to obtain 2', 3'-cyclic phosphoric acid body, hydrolyzing the same with an alkali such as sodium hydroxide to form 2'(3')-phosphoric body then performing reduction of the azide group to an amino group with, for example, hydrogen sulfide-(pyridine-water). A polysaccharide gel such as agarose or crosslinked dextran and a synthetic polymer gel such as polyvinyl gel having a hydroxyl group are used as the above-mentioned base having the hydroxyl group. B in the formula denotes a nucleide base, R<1> denotes a hydrogen atom, hydroxyl group or phosphoric acid group and R<2> denotes a hydroxyl group or phosphoric acid group.
