Simple, cheap, rapid extn. of the cardiotonic agent cinobufagin by (a) extn. of finely powdered Bufonis venenum with ethyl acetate for >=2 hrs., pref. at 50 degrees C and filtering; (b) evaporating the filtrate from (a) and extracting the residue for >=2 hrs.; (c) concentrating the extract, filtering off the sterols and evaporating the filtrate; (d) dissolving the residue from (c) in ethyl acetate and chromatographing the soln. on moist silica gel, eluting with ethyl acetate and evaporating the eluate; (e) sepg. cinobufagin by chromatographing the residue from (d) on dry silica gel, eluting with acetone/hexane (1:5) and (f) evaporating the eluate. Cinobufagin and resibufogenin are the main components of the residue from (d) and occur in the first fractions. Cinobufagin occurs in the first fractions of stage (e). "Bufonis venenum" is a crude, crystalline endocrine secretion from the common toad. |
