PURPOSE:To achieve the fusion of protoplast in high frequency, by carrying out the fusion of bacterial protoplast in a hypertonic solution containing polyethylene glycol and calcium ions at 8-12pH. CONSTITUTION:The protoplast of bacterium belonging to Brevibacterium genus or Corynebacterium genus is mixed with the protoplast of bacterium belonging to Bacillus genus, centrifuged, and suspended in an alkaline hypertonic solution of 8.0-12.0pH containing >=10mM, preferably 10-100mM of calcium ions. A solution of polyethylene glycol is added to the suspension, and the cell fusion is induced in the presence of >=10mM of calcium ions and 10-50% polyethylene glycol at 8-12pH. |
