NEW MATERIAL:A cyclic plasmid obtained from Clostridium butyricum, having a molecular weight of 51 1 M-dalton and decomposable to 10 fragments by the restriction enzyme EcoRI, 7 fragments by SalI and 8 fragment by BamHI; and a cyclic plasmid having a molecular weight of 32 1 M-dalton, and decomposable to 5 fragments by DcoRI and undecomposable by SalI, BamHI and PstI. USE:A vector for the gene recombination using a bacterium belonging to clostridia genus as a host. PROCESS:Clostridium butyricum IFO3857 strain belonging to clostridia genus is cultured, and cleared lysate (supernatant liquid) is prepared from the cultured liquid. The cyclic plasmid is prepared from the liquid by centrifugation using cesium chloride-ethidium bromide. |
