PURPOSE:To prepare a peptide by protecting the imidazole group of an imidazole group-containing compound with a specific group, subjecting the compound to peptide condensation, and removing the protecting group. CONSTITUTION:A peptide having imidazole group is prepared by (1) protecting the imidazole group of an imidazole group-containing compound (e.g. histidine) with 4-methoxy-2,3,6-trimethylbenzenesulfonyl group, (2) subjecting the product to peptide condensation, and (3) removing the protecting group with an acid (e.g. trifluoroacetic acid, etc.) or 1-hydroxybenzotriazole, especially in the presence of thioanisole, anisole or dimethylsulfide. The protecting group is stable to catalytic reduction and an acid such as trifluoroacetic acid, and also to a base such as triethylamine, etc. |
